Feasibility of synchrotron radiation computed tomography on rats bearing glioma after iodine or gadolinium injection

Abstract: The purpose of this work was to demonstrate the feasibility of a new imaging technique called synchrotron radiation computed tomography (SRCT). This technique leads to a direct assessment of the in vivo concentration of an iodine- or gadolinium-labeled compound. Rats bearing C6 glioma were imaged by MRI prior to the SRCT experiment. The SRCT experiments were performed after a 1.3 g I/kg (n = 5) or a 0.4 g Gd/kg (n = 5) injection. Finally, brains were sampled for histology. The SRCT images exhibited contrast en… Show more

“…C6 gliomas were originally produced as a result of exposing Wistar-furth rats to N-nitrosomethylurea, and then isolated and grown as a cell culture [ 59 ]. This tumour model has been used in multiple studies involving conventional radiotherapy [ 60 62 ] and synchrotron radiation [ 63 65 ].…”

“…This method allowed for detailed spatial analysis of BBB damage, while allowing us to directly relate BBB permeability to PHZ ion dyshomeostasis. We are the first to use this approach in rodent stroke models, although others have measured Gd with synchrotron techniques to assess other pathologies [ 29 , 30 ]. XFI was compared to ICP-MS.…”

“…Monochromatic imaging allows elemental decomposition at a higher accuracy than state-of-the-art clinical imaging 13 . In addition, the absolute concentration of the contrast agent 2830 can be determined in KES CT as well as the electron density of the material after performing a dual-energy approach 3133 . In most KES CT synchrotron studies the clinical applicability was limited by the dose applied to the specimen, with dose levels between 7 mGy 33 and 9 Gy (skin entry dose) 19 .…”
