Febuxostat ameliorates high salt intake-induced hypertension and renal damage in Dahl salt-sensitive rats

Abstract: Objective: Several clinical studies have reported that xanthine oxidoreductase inhibitors have antihypertensive and renal protective effects but their mechanisms have not been fully determined. This study aims to clarify these mechanisms by examining the effects of febuxostat, which is a novel selective xanthine oxidoreductase inhibitor, in Dahl salt-sensitive rats. Methods: Eight-week-old male Dahl salt-sensitive rats were fed a normal salt (0.6% NaCl) or high salt (8% NaCl) diet for 8 weeks. A portion of the … Show more

“…Febuxostat attenuated proteinuria, renal dysfunction, and renal interstitial fibrosis but did not affect blood pressure in Nx-induced CRF rats [16]. Febuxostat attenuated hypertension, renal dysfunction, and renal histology in Dahl salt-sensitive rats fed a high-salt diet [17]. These results suggest that the ROS generation by NADPH oxidase and XO may induce blood pressure elevation and renal damages, respectively, and that superoxide dismutase may not affect blood pressure or renal damage in Nx-induced CRF rats.…”

“…Febuxostat is considered a selective inhibitor of xanthine oxidoreductase (XOR), an enzyme that catalyzes the formation of uric acid. In Dahl salt-sensitive rats, orally administered febuxostat decreased the urinary excretion of TBARS, which was measured by means of a commercially available colorimetric batch TBARS assay [76]. XOR is also considered to generate superoxide anions, which in turn can mediate lipid peroxidation and increase MDA formation.…”
