Femtosecond spectroscopic study of carminic acid–DNA interactions

Abstract: Photo-excited carminic acid and carminic acid-DNA complexes in a buffer solution at pH= 7 have been examined using a variety of spectroscopy techniques, that are in particular, the femtosecond resolved fluorescence up-conversion and transient absorption spectroscopy M -1 that is typical for anthracycline-DNA complexes.

“…Deconvolution of the emission spectrum also results in two distinct Gaussian components at 16 180 (Δν 1/2 = 2140 cm −1 ) and 17 070 cm −1 (Δν 1/2 = 1170 cm −1 ), which can be attributed to LE and ICT emissions, respectively (Figure a). Weak band at 14200 cm −1 can be attributed to the traces of carminic acid monoanion. , …”

“…It was also used recently for preparation of red silica glasses using sol−gel method . Because of its specific interactions with proteins, nucleic acids, and lipids, carminic acid also found numerous applications in biochemical studies. Structure of the carminic acid molecule suggests strong covalent binding via carboxylic and/or hydroxyl functionalities.…”

“…Although CarOH cannot bind to DNA,5 it can suppress the genotoxic activities of several mutagens in the Drosophila DNA repair test and in the Ames test 6. Recently, CarOH is found to be a ligand to form a complex with DNA and to be a molecular probe in femtosecond spectroscopic study on DNA 7. CarOH can produce superoxide anion when it is oxidized by Agaricus bisporus tyrosinase 8.…”

“…Physical analysis methods, generally non-destructive, as-sume the interaction of the substance with radiation of a certain energy. The optical properties of organic molecules can be examined using stationary techniques such as spectroscopy that is a method based on the property of substances to selectively absorb electromagnetic radiation, therefore in the last several years, it has become possible to directly observe the dynamics of excited electronic states in DNA model compounds by femtosecond spectroscopy [8]. In the case of the interaction of radiation with the substance, transitions are present due to the absorption of energy by the molecules.…”
