“…The high concentration of 2,4-D (7 mg l -1 ) used for subculture could shorten the time required for embryogenic callus selection, an observation made by Wang et al (1990) for wheat. The suitability of the amino acid-based culture media AA for the rapid establishment of a rice suspension culture that readily yields protoplasts has been noted in previous studies Jenes and Pauk 1989;Bhattacharjee and Gupta 1995;Jain et al 1995;Xue and Earle 1995). In the present investigation, suspensions were successfully established in both AA medium and MS-based mMS medium.…”

“…In our experiment, the optimal plating density with the highest division frequency and plating efficiency was 110 6 protoplasts per milliliter medium, and a lower or higher density resulted in decreased division frequency and plating efficiency (Table 2). A similar density was used in barley (J盲hne et al 1991;Funatsuki et al 1992) and rice (Bhattacharjee and Gupta 1995;Xue and Earle 1995;Fujimura et al 1985). KM8p medium was used for protoplast culture, and of the four hormone and sugar solutions tested in the medium, solution 1 proved to be the most effective for cell division frequency and plating efficiency (Table 3).…”
