Ferulic acid promoting apoptosis in human osteosarcoma cell lines

Abstract: Objective:To explore the promoting apoptosis and antitumor activities of ferulic acid (FA) in human osteosarcoma and its potential mechanism. Methods:The SaOS-2 and MG63 osteosarcoma cell lines were opted to experiment and these cells were, respectively, cultured with various concentrations of FA (0 μM, 10 μM, 20 μM, 40 μM) for 72 hours at 37°C. The viabilities of the FA treated cells were monitored by MTT. Apoptosis cells were evaluated using annexin V/PI by flow cytometry. Apoptosis proteins caspase-3, procas… Show more

“…Furthermore, FA inhibited proliferation and induced apoptosis in 143B and MG63 osteosarcoma cells dose-dependently leading to G0/G1 phase arrest and apoptosis by down-regulating the expression of CDK 2, 4 and 6, upregulating Bax, downregulating Bcl-2, subsequently enhancing caspase-3 activity. It also inhibited PI3K/Akt activation in both cell lines [73,74]. FA role in suppressing metastasis was regulated by the reversal of epithelial-mesenchymal transition in vitro in MDA-MB-231 moreover, the inhibition of cell invasion through reducing MMP-9 mRNA expression [75].…”

“…In the previous studies, FA is an effective antioxidant agent that protects DNA from oxidative damage and prevents lipid peroxidation through reducing oxidative stress [ 11 ]. In many tumor cell lines such as human osteosarcoma, human glioblastoma (U87MG), and prostate cancer, FA can induce cytotoxicity [ 12 14 ]. Due to the inhibition of cyclooxygenase-2, FA is considered to be an anti-proliferative agent [ 15 ].…”

“…Ferulic acid, as the major phenolic acid in the plant extract, is reported for its proapoptotic and antiproliferative activities against various cancer cell lines [20,21]. Rosmarinic acid was also reported to induce cell cycle arrest and apoptosis on many cancer cell lines [22,23].…”
