FGFR3 Down-Regulation is Involved in bacillus Calmette-Guérin Induced Bladder Tumor Growth Inhibition

Abstract: Based on bladder cancer murine model results we infer that down-regulation of FGFR3 is a predictive marker of a good response to bacillus Calmette-Guérin therapy. The decrease in FGFR3 in response to bacillus Calmette-Guérin occurred not only in a murine model but also in a human bladder cancer cell line and in some patient samples. More patients and increased followup are needed to establish the predictive role of FGFR3 as a marker in human bladder cancer.

“…We investigated the response to BCG therapy. Expression of FGFR3 has been found a BCG response marker, which corroborates the data in mice from Langle et al 21 . Indeed, in the Langle et al study, the responder group showed low expression of FGFR3, which we also found.…”

“…We also found that FGFR3, a member of the bladder cancer signaling pathway, was significantly downregulated, indicating an aggressive characteristic and a worse prognosis. In the aspect of FGFR3 expression, those expression levels in the EP subtype were significantly higher than in the others across NMIBC patient cohorts (Supplementary Figure S2), supporting a poor responsiveness to BCG treatment in the EP subtype and consistent with the previous investigation [18]. The alteration in expression levels of genes involved in REC.BCG+-associated pathways was validated in the UROMOL cohort 5A).…”

“…42 FGFR3 is a receptor tyrosine kinase that is involved in cell functions such as proliferation, differentiation, and development of tumor cells. 43 These results suggest that BAPN-1 may potentially affect targets that are essential for cancer survival. Additionally, we observed that the upregulated genes upon BAPN-1 treatment were mainly related to inflammatory and immunomodulatory responses.…”
