Fibrinogen is required for maintenance of platelet intracellular and cell-surface P-selectin expression

Abstract: Platelet P-selectin plays important roles in inflammation and contributes to thrombosis and hemostasis. Although it has been reported that von Willebrand factor (VWF) affects P-selectin expression on endothelial cells, little information is available regarding regulation of platelet P-selectin expression. Here, we first observed that P-selectin expression was significantly decreased on platelets of fibrinogen and VWF double-deficient mice. Subsequently, we identified this was due to fibrinogen deficiency. Impa… Show more

“…Interestingly, our data also suggest that platelet LRP8 is not required for leukocyte adhesion, although platelets have been reported to contribute to leukocyte rolling. 51,52 It is noteworthy that LRP8 Ϫ/Ϫ mice do not experience hypercholesterolemia as LDLR knockout mice do 53 ; thus, the HNP-induced platelet activation is likely independent of modification of lipid metabolism, although this issue is not addressed in the present study. Furthermore, LRP8…”

“…Preparation of plasma, platelets, and pFn. Mouse plasma and platelets were prepared as previously described (18,22,25,(50)(51)(52). For pFn-fibrin clot formation in vitro, blood was drawn from inferior vena cava with a 25-gauge needle and immediately mixed with 3.2%…”

“…He was treated with corticosteroids with complete response. He experienced 3 additional episodes of thrombocytopenia four, two, and one year before the current admission (platelet count nadirs were 35x10 9 /L, 13x10 9 /L, and 1x10 9 /L, respectively), each treated with corticosteroids as an outpatient with normal platelet counts between relapses. Relapses were not associated with hemorrhagic symptoms other than petechiae.…”
