Fibronectin adsorption, conformation, and orientation on polystyrene substrates studied by radiolabeling, XPS, and ToF SIMS

Abstract: Protein adsorption is widely studied by a variety of techniques, but there still is little known about protein orientation and conformation after adsorption. This probably is due to the large number of parameters involved, such as the characteristics of the surface and the structure of the protein. In this study, the adsorption of fibronectin was investigated with three different techniques: radiolabeling, X-ray photoelectron spectroscopy (XPS), and time-of-flight secondary ion mass spectrometry (ToF SIMS) on … Show more

“…On the other hand, super-hydrophilic surface of titanium dioxide films 33 or O 2 -treated HMDSO films that showed a high rate of cell attachment had a rigid surface. It is also necessary to investigate the protein conformation that influences cell adhesion by using a TOF-SIMS system 29,30,34 for better understanding of these phenomena.…”

“…This is one of the bottleneck problems in the studies of protein adsorption. At present, there are many methods used for the measurements of protein adsorption, including radio-labeling [12] , antibody techniques [13,14] , QCM [15] , AFM [16,17] , the second mass spectra [18] , photoelectron spectroscopy analysis [19] , etc. However, until now, no any singular technology can overall analyze the protein adsorption process, including the categories and quantity of the adsorbed proteins, the conformational change after adsorption, and particularly the quantitative analysis of secondary structural change after adsorption.…”

“…The statistical method for the analysis of TOF-SIMS data was informed by the papers Lhoest et al (24,25) and Wagner et al (38,39). A statistical analysis was conducted to identify the molecules adsorbing from BALF to PM2.5.…”
