Field-Usable Lateral Flow Immunoassay for the Rapid Detection of White Spot Syndrome Virus (WSSV)

Abstract: BackgroundWhite spot disease (WSD), a major threat to sustainable aquaculture worldwide, is caused by White spot syndrome virus (WSSV). The diagnosis of WSD relies heavily on molecular detection of the virus by one-step PCR. These procedures are neither field-usable nor rapid enough considering the speed at which the virus spreads. Thus, development of a rapid, reliable and field-usable diagnostic method for the detection of WSSV infection is imperative to prevent huge economic losses.Methods/Principal Finding… Show more

“…Due to this simplicity, LFIAs have been used in a variety of settings, including clinical, food safety, and environmental analyses. Compared to standard laboratory technologies, LFIAs are simple-to-use, rapid, low-cost, and portable, thus meeting the criteria for healthcare in resource-limited settings 1 , 2 , and have been widely used for the detection of various targets, such as tumor markers 3 , 4 , bacterium 5 , viruses 6 , nucleic acids 7 , and pesticide residues 8 . However, poor sensitivity limits the further application of this testing platform 9 .…”

“…Meanwhile, sandwich immunoassay test kits were also developed recently141516. Unfortunately, all these molecular techniques have their own limitations such as; less sensitivity and high detection limit (~10 3 copies of its gene μL −1 ), complicated offline preparation procedures, involvement of carcinogenic chemicals like ethidium bromide (in polymerase chain reaction (PCR)), requirement for well-equipped laboratories (for DNA assays), trained technicians and time consuming measurements (For ELISA 2–3 days’ of time period required) and limited to qualitative assay (ELISA kit).…”

“…Accumulation of bound particles makes the band visible to the naked eye and is indicative of a positive reaction (Posthuma-Trumple et al, 2009;De Boer and Lopez, 2012). The second capture line is the control line, containing species-specific antibodies designed to capture all unbound tagged antibodies that would also become visible to confirm that the assay has run correctly (Kulabhusan et al, 2017).…”
