Firefly luciferase gene: structure and expression in mammalian cells.

Abstract: The nucleotide sequence of the luciferase gene from the firefly Photinus pyralis was determined from the analysis of cDNA and genomic clones. The gene contains six introns, all less than 60 bases in length. The 5' end of the luciferase mRNA was determined by both Si nuclease analysis and primer extension. Although the luciferase cDNA clone lacked the six N-terminal codons of the open reading frame, we were able to reconstruct the equivalent of a full-length cDNA using the genomic clone as a source of the missi… Show more

“…After 48 h cell extracts were prepared in lysis bu er (10 mM HEPES pH 7.9, 400 mM NaCl, 0.1 mM EGTA, 0.5 mM DTT, 5% glycerol, 0.5 mM PMSF) and then chloramphenicol acetyltransferase and luciferase activities were determined (de Wet et al, 1987;Gorman et al, 1982).…”

“…The pMTI-Luc vector is a modified version of the luciferase reporter construct pSV2A, 31 in which the SV40 promoter has been replaced with the MT-I promoter. The fragments were cloned by standard blunt-end cloning protocols in the sense (*ÂŒS) and antisense (*ÂŒA) direction immediately upstream of the mouse MT-I promoter into the BamHI site of this vector (pMTB*-2, -4 and -7), or in the SmaI site 139 bp downstream from the stop codon of the luciferase gene (pMTS*-2, -4 and -7).…”

“…Luciferase gene expression was measured by luminescence according to de Wet et al 31 The luminescence generated upon automatic addition of 150 l of 167 M luciferin dissolved in water was recorded in duplicate samples for 4 s by a luminometer (Lumat LB 9501; Berthold, Wildbach, Germany). The luminescence is reported as relative light units (RLU).…”
