First report and new molecular and morphological characterizations of root-knot nematode, Meloidogyne javanica, infecting ginger and long coriander in Vietnam

Abstract: Ginger (Zingiber officinale Roscoe) and long coriander (Eryngium foetidum L.) are commonly grown and used as important spices and medicinal plants in Vietnam. Our study recovered for the first time one of the most damaging tropical root-knot nematodes, Meloidogyne javanica, associated with these plants in the Western Highlands of Vietnam. In this study, M. javanica was characterized based on morphology and molecular characterization of D2-D3 fragment of 28S rRNA, ITS, and Nad5 mtDNA regions. The identification… Show more

“…Juvenil 2 nematodes Meloidogyne sp. A vermiform elongated body shape characterized those found in this study, having a slender straight stylet with a small round basal knob, oval and well-developed metacorpus, an overlapping esophagus with a pointed and serrated tail end intestine, and hyaline at the tail end [13]. The population of nematodes female Pratylenchus sp.…”

“…The classification up to the generic level of PPNs was based on Decraemer and Hunt (2013). To confirm morphological identification, nuclear (18S rDNA, internal transcribed spacer [ITS] and 28S rDNA) and mitochondrial ( COI , COII /16S rDNA, and Nad5 ) gene regions of nematodes were amplified by PCR and analysed where possible (Janssen et al, 2016; Nguyen et al, 2022; Nguyen, Nguyen, et al, 2021; Phan et al, 2020). To identify tropical Meloidogyne spp., a multiplex PCR with species‐specific primers has also been employed (Kiewnick et al, 2013).…”

“…After 1 week of fixing, nematodes were transferred to anhydrous glycerine to make permanent slides, following Seinhorst (1959). Perineal patterns of adult Meloidogyne females were mounted in glycerine following Hartman and Sasser (1985) were amplified by PCR and analysed where possible (Janssen et al, 2016;Nguyen et al, 2022;Phan et al, 2020).
