First step towards the biomolecular characterization of Pompia, an endemicCitrus-like fruit from Sardinia (Italy)

Abstract: This study is the first molecular and biochemical analysis conducted on Pompia, a plant of unknown origin that is endemic to Sardinia; this plant is thought to belong to the Citrus genus. Here, genes coding for the enzymes superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), peroxidase (POD, EC 1.11.1.7), and polyphenol oxidase (PPO, EC 1.14.18.1) were identified. We detected the aforementioned enzymes in fresh leaf tissue and assessed the catalytic activity of each to support the molecular an… Show more

“…A degenerate hybrid oligonucleotide primer (CODEHOP) strategy [ 48 ] was used to detect unknown nucleotide sequences of the CC genes. The CODEHOP strategy has proven to be useful for the exploration of large gene families in plants [ 49 ] and for the isolation of different plant genes related to antioxidant activity [ 50 , 51 , 52 , 53 ]. According to the CODEHOP strategy, we selected six PPO amino acidic sequences of different plant sources from the GenBank SwissProt database.…”

“…Our body can counteract the activity of free radicals through endogenous antioxidant mechanisms and the introduction through the diet of exogenous substances [89]. Among the endogenous factors are enzymes such as superoxide dismutase (SOD; EC 1.15.1.1) which catalyzes the dismutation of the superoxide radical (O 2 − ) into either molecular oxygen (O 2 ) or hydrogen peroxide (H 2 O 2 ), catalase (CAT; EC 1.11.1.6), which removes intracellular H 2 O 2 , and reduced glutathione (GSH) [90]. Among the non-exogenous non-enzymatic substances with antioxidant properties, we recall Vitamin E, Vitamin C, carotenoids, polyphenols, and anthocyanins instead.…”
