BackgroundThe continuous emergence of multi-drug-resistant bacteria drastically reduces the efficacy of antibiotic armory and, consequently, increases the frequency of therapeutic failure. The discovery of new antibacterial drugs is an urgent need. The present study reports the antibacterial and antioxidant activities of the methanol extract, fractions and iridoids from Canthium subcordatum, a plant traditionally used as antidiabetic, anti-inflammatory, and antimicrobial.MethodsBroth microdilution assay was used to determine minimum inhibitory concentrations (MICs) and minimum bactericidal concentrations (MBCs) of extracts and iridoids against Staphylococcus aureus, Vibrio cholerae and Shigella flexneri. Antioxidant activity was evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and gallic acid equivalent antioxidant capacity (GAEAC) assays. The samples were also tested for their cytotoxicity against human red blood cells (RBC).ResultsThe methanol extract, hexane, ethyl acetate and iso-butanol fractions from C. subcordatum fruits displayed different degrees of antioxidant (EC50 = 62.83–70.17 μg/ml; GAEAC = 45.63–58.23 μg/ml) and antibacterial (MIC = 128–512 μg/ml) activities. Canthiumoside 1(1) and linearin (7) were the most active antioxidant (EC50 = 1.12–2.03 μg/ml; GAEAC = 79.82–92.35 μg/ml) and antibacterial (MIC = 8–64 μg/ml) compounds while the most sensitive bacterium was Staphylococcus aureus. The tested samples were non-toxic to normal cells.ConclusionOur results demonstrated that compounds 1 and 7 were potent antibacterial agents and DPPH/ABTS·+ radical scavengers, so they warrant further investigation.

Characterization and Antimicrobial Activity of Volatile Constituents from Fresh Fruits of Alchornea cordifolia and Canthium subcordatum

Bacterial resistance has been increasingly reported worldwide and is one of the major causes of failure in the treatment of infectious diseases. Natural-based products, including plant secondary metabolites (phytochemicals), can be exploited to ameliorate the problem of microbial resistance. The fruit essential oils of Alchornea cordifolia and Canthium subcordatum were obtained by hydrodistillation and analyzed by gas chromatography-mass spectrometry (GC-MS). The essential oils were subjected to in vitro antibacterial, antifungal and cytotoxic activity screening. Thirty-eight compounds comprising 97.7% of A. cordifolia oil and forty-six constituents representing 98.2% of C. subcordatum oil were identified. The major components in A. cordifolia oil were methyl salicylate (25.3%), citronellol (21.4%), α-phellandrene (7.4%), terpinolene (5.7%) and 1,8-cineole (5.5%). Benzaldehyde (28.0%), β-caryophyllene (15.5%), (E,E)-α-farnesene (5.3%) and methyl salicylate (4.5%) were the quantitatively significant constituents in C. subcordatum fruit essential oil. A. cordifolia essential oil demonstrated potent in vitro antibacterial activity against Staphylococcus aureus (MIC = 78 μg/mL) and marginal antifungal activity against Aspergillus niger (MIC = 156 μg/mL). C. subcordatum showed antibacterial activity against Bacillus cereus and S. aureus (MIC = 156 μg/mL) and notable antifungal activity against A. niger (MIC = 39 μg/mL). However, no appreciable cytotoxic effects on human breast carcinoma cells (Hs 578T) and human prostate carcinoma cells (PC-3) were observed for either essential oil. The antimicrobial activities of A. cordifolia and C. subcordatum fruit essential oils are a function of their distinct chemical profiles; their volatiles and biological activities are reported for the first time.

Linking phytochemistry to traditional uses and pharmacology of an underexplored genus – Psydrax: a review

The genus Psydrax is one of the ethno-medicinally important genera of the Rubiaceae family which has only received a limited scientific attention, despite coming from a pharmacologically and phytochemically important plant family. The genus has found applications in ethnomedical management of diabetes, stomach disorders, inflammations, cardiovascular diseases, epilepsy, wounds, malaria and fever. To unveil knowledge gaps, stimulate research interest and unravel opportunities for drug discovery from the genus Psydrax , we have carried out an extensive review on its traditional applications, phytochemistry and pharmacology for the first time. Literature on these topics was obtained from Google Scholar, Pubmed and ScienceDirect journal articles published from 1788 to September, 2021. Only articles written in English were reviewed. While several species of Pysdrax used in traditional medicine have not been chemically explored for drug discovery, over a hundred secondary metabolites have so far been identified in few species of the genus, and majority of these chemotaxonomic markers are iridoids. Bioactive extracts and some isolated constituents of Psydrax species have shown various in vitro and in vivo pharmacological properties including anti-hyperglycemia, anti-inflammatory, anticonvulsant and antimicrobial, and thus, support some of the ethnomedical uses of the plants. For an evidence-informed application of the genus, Psydrax , in traditional medicine, more ethnobotanical surveys, elaborate in vivo pharmacological assays, in-depth toxicity and holistic phytochemical studies are required to fully exploit more species of the genus prior to future clinical studies. Following documented traditional uses of Psydrax species, the deliberate cultivation of medicinal plants under this genus is recommended for sustainability in medicinal plant utilization.
