Five omic technologies are concordant in differentiating the biochemical characteristics of the berries of five grapevine (Vitis vinifera L.) cultivars

Abstract: BackgroundGrape cultivars and wines are distinguishable by their color, flavor and aroma profiles. Omic analyses (transcripts, proteins and metabolites) are powerful tools for assessing biochemical differences in biological systems.ResultsBerry skins of red- (Cabernet Sauvignon, Merlot, Pinot Noir) and white-skinned (Chardonnay, Semillon) wine grapes were harvested near optimum maturity (°Brix-to-titratable acidity ratio) from the same experimental vineyard. The cultivars were exposed to a mild, seasonal water… Show more

“…In RNO, the total RNA was extracted from berry skins at 20, 22, 24 and 26°Brix (three experimental replicates each), and the quantity and quality was assessed, as previously described [109] from approximately 250 mg of frozen, finely ground, skin tissue.…”

“…were used for sequencing. In RNO, total RNA was extracted from 21 approximately 250 mg of frozen, finely ground, skin tissue using a modified 22 CTAB extraction protocol followed by an additional on-column DNase 23 digestion using a Qiagen RNeasy Mini Kit (Qiagen, Valencia, CA, USA) as in 24 [109]. RNA quality and quantity were assessed with a Nanodrop ND-1000 25 spectrophotometer (ThermoFisher Scientific, Waltham, MA, USA) and an 1 Agilent 2100 Bioanalyzer (Agilent Technologies, Santa Clara, CA, USA).…”

“…A number of molecular biology tools have been developed to study a range of biological problems. Proteomics has been proved an effective tool to analyze the proteome changes in grape under abiotic and biotic stress as well as during berry development 23 27 . Dimensional electrophoresis followed by Matrix-Assisted Laser Desorption/Ionization Time of Flight Mass Spectrometry (MALDI-TOFI-MS) has been used to characterize the differential expressed proteins in V. vinifera 28 31 .…”
