Flaviviridae virus nonstructural proteins 5 and 5A mediate viral immune evasion and are promising targets in drug development

Abstract: Infections with viruses in the Flaviviridae family have a vast global and economic impact because of the high morbidity and mortality. The pathogenesis of Flaviviridae infections is very complex and not fully understood because these viruses can inhibit multiple immune pathways including the complement system, NK cells, and IFN induction and signalling pathways. The non-structural (NS) 5 and 5A proteins of Flaviviridae viruses are highly conserved and play an important role in resisting host immunity through v… Show more

“…The ability of pathogenic viral proteins to interfere with the innate and adaptive immune response in the form of altered gene expression conferring tolerance or other forms of immune evasion has been observed. 120,121 For the coronaviruses, several structural and non-structural proteins which play a role in pathways leading to IFN-I and III expression have been identified, 122 including nsp16, 123 membrane, 124,125 nucleocapsid, 126 amongst others. Some of these immune evasive proteins have a high homology with SARS-CoV-2 proteins.…”

“…Flaviviral NS5 interacts with different host proteins in an infected cell. Some of these interactions are relevant to the process of viral RNA genome synthesis, while in some other cases the interaction of NS5 with other host factors can interfere with or modulate cellular functions [19].…”

“…Unlike DENV, YFV NS4B has also been shown to block RIG-I stimulation through an interaction with STING (32). Moreover, NS5 is known to establish a common mode of IFNa/b signaling antagonist for some flaviviruses by inhibiting the JAK/STAT pathway (33,34).…”
