FleQ regulates both the type VI secretion system and flagella in Pseudomonas putida

Abstract: Based on the analysis of sodium dodecyl sulfate-polyacrylamide gel electrophoresis and mass spectrometry, flagellin FliC and a key member of the type VI secretion system Hcp1 were found to be secreted from Pseudomonas putida KT2442 cells in the late exponential phase, but in the early exponential phase only FliC was secreted. Relevant genes fleQ and rpoN were then individually deleted in P. putida KT2442, and the expression and transcription of fliC and hcp1 in these mutant strains were analyzed. In both △fleQ… Show more

“…FleQ is the most important transcription factor (TF) and master regulator of biofilm formation and motility responsive to cdGMP. The same TF is also related to other functions, including iron homeostasis (Blanco-Romero et al, 2018) and the control of Type 6 secretion system (Wang et al, 2018). In P. putida in conditions of low cdGMP, FleQ can interact with σ 54 to regulate a high number of promoters related to motility and flagella.…”

“…In this regulatory cascade, the transcriptional regulator FleQ appears to be the master regulator 2 . The function of FleQ in flagella synthesis regulation has been studied in Pseudomonas aeruginosa 1 , 2 , in P. putida 3 5 and in P. fluorescens 6 8 . In these species, mutations in the fleQ gene result in non-motile, aflagellated bacteria.…”

“…(1234567890) www.nature.com/scientificreports/ and it has been previously linked to the control of T6SS in P. aeruginosa 52 . FleQ belongs to the NtrC/NifA family and has been related to the regulation of T6SS and c-di-GMP metabolism in P. putida 89 . The differential gene expression analyses shown in Fig.…”
