Forearm beta adrenergic receptor-mediated vasodilation is impaired, without alteration of forearm norepinephrine spillover, in borderline hypertension.

Abstract: Impaired beta adrenoceptor-mediated vasodilation associated with enhanced sympathetic activity has been reported in established hypertension. We examined whether altered beta adrenoceptor-mediated vasodilation occurs early in the disease process, when structural vascular changes are likely to be less marked, by measurement of forearm blood flow by strain gauge plethysmography after the intraarterial administration of increasing doses of a beta receptor agonist, isoproterenol, in eight subjects with borderline … Show more

“…Conversely, the attenuated response to dobutamine might result from HT-induced impairment in ␤-adrenoceptor-mediated vasodilation 40,41 or might reflect vascular remodeling. The greater improvement in the responses to adenosine than to dobutamine challenge in vitamin-treated HT might at least in part be related to their different mechanisms of action, because antioxidants are known to preferentially improve endothelium-dependent function.…”

“…Upstream of its coding region is the coding region for a 19 amino acid peptide, termed 5′ leader cistron, that has been implicated in receptor expression. Since blunted β-adrenoceptor-mediated vasodilation has been observed in hypertension, the β2AR has become a candidate gene for this disease (Stein et al 1995). To date, several polymorphisms have been identified in the β2AR, five of which change the primary sequence (Fig.…”

“…Indeed, in hypertensive conditions, several reports indicate a reduction in ␤AR signaling and regulation. [5][6][7][8][9][10] If these premises hold true, improving ␤AR signaling should result in the restoration of ␤AR vasorelaxation. In fact, some interventions have been effective in correcting ␤AR signaling in hypertension, such as dietary salt restriction 7 or pharmacological treatment.…”
