Forkhead box C1 boosts triple‐negative breast cancer metastasis through activating the transcription of chemokine receptor‐4

Abstract: The transcription factor forkhead box C1 (FOXC1) has recently been proposed as a crucial regulator of triple‐negative breast cancer (TNBC) and associated with TNBC metastasis. However, the mechanism of FOXC1 in TNBC development and metastasis is elusive. In this study, overexpression of FOXC1 in MDA‐MB‐231 cells significantly enhanced, whereas knockdown of FOXC1 in BT549 cells significantly reduced, the capabilities of TNBC cell invasion and motility in vitro and metastasis to the lung in vivo, when compared t… Show more

“…Two key genes of TNBC, ER and PR, were among the genes selected by enetLTS. Other genes selected by enetLTS, FOXA1 [ 25 ], GATA3 [ 25 ], SPDEF [ 26 ], FOXC1 [ 27 ], EN1 [ 28 ], HORMAD1 [ 29 ], KRT16 [ 30 ], and CT83 [ 31 ], have been reported to be related to TNBC.…”

“…Moreover, patient data analysis shows that overexpression of KLF8 and CXCR4 correlates with metastatic potential [ 38 ]. Likewise, transcription factor forkhead box C1 (FOXC1) induces the expression of CXCR4 and enhances migration of MDA-MD-231 cells and lung metastasis in vivo [ 39 ]. In addition, POU domain class 2 transcription factor 1 (known as POU1F1 (Pit-1)) induces the expression of both CXCR4 and CXCL12 in breast cancer cells [ 40 ].…”

“…The metastatic potential of triple-negative breast cancer (TNBC) to lungs has been found to be regulated by another transcription factor, forkhead box C1 (FOXC1). This protein, by increasing the expression of chemokine receptor-4 (CXCR4), also promotes the Lung metastatic capability of Breast cancer cells in vivo [ 92 ]. In conclusion, Snail 1 and forkhead box C1 that influence the immune microenvironment in Breast cancer, if targeted therapeutically, the desired results to overcome breast cancer lung metastasis in the future, can, therefore, be accomplished.…”
