Objective: The present research work is focused to develop in situ raft gel of Nizatidine. sodium alginate (SA) is one of the critical components for the development in situ raft system. Methods: The formulation was prepared using polymers such as SA and gellan gum. The formulations were subjected to evaluation characteristics such as pH, in vitro gelling capacity, viscosity, gel strength, and in vitro release studies. Results: The pH of all the prepared batches was found in the range of 6.4–7.2 for S1–S9, and 6.3–7.2 for G1–G9. S1–S9 formulations showed viscosity in the range of 253.7–400.9 cps, and G1–G9 formulations showed viscosity in the range of 253.4–399.8 cps. Formulation S6 containing SA and G6 containing gellan gum gave the highest drug content of 99.58% and 99.5%, respectively. The highest gel strength 4.6 is exhibited by S6 and G6 formulations. Formulation S6 containing SA gave the highest drug release of 95.62% and also showed sustained and controlled release for up to 12 h. Conclusion: Gastric raft formulation is a better choice for drugs such as Nizatidine which enhances the drug release for a prolonged time by remaining buoyant in the stomach for more than 12 h.

Lisinopril is a potent ACE (angiotensin-converting enzyme) inhibitor used to treat hypertension and congestive heart failure. It exhibits 25% of low bioavailability. Hence, in the current study, the major objective was to increase the gastric transient time of lisinopril and develop in situ gel formulation for better absorption and modulating release behavior of lisinopril. Different formulations of lisinopril were prepared by using gelling polymers such as Carboxymethyl cellulose (CMC), pectin, and calcium carbonate. Sodium citrate was used to prevent gelation outside the gastric environment. The formulation was studied using Fourier Transform Infrared Spectroscopy (FTIR) and Differential Scanning Calorimetry (DSC) to interpret the interaction between drugsand polymers. For optimization of in situ gelling system 3 2 full factorial design was employed to study the effect of independent variables, the concentration of CMC (X1) and concentration of sodium alginate (X2), on the dependent variables viscosity, and % drug content. Formulation (F5) containing 1.25% of sodium alginate, and 0.75% of CMC showed good gelling ability. The composition F5 was optimized on the basis of viscosity (5.03 Pa.s), drug content (94.06±1.0%), and cumulative drug release (95±0.73%) at 12 h. Floating in situ gelling system improved bioavailability and gastric transit time of lisinopril. A stability study indicated the absence of any noticeable change in the formulation. Thus, in situ gel formulation is a promising approach for gastro-retentive sustained delivery of lisinopril. These results ensure that the developed system is an alternative to conventional drug delivery systems and can enhance patient compliance.

Objective: The current research involves the formulation of a sustained-release gastro-retentive drug delivery system of nizatidine. Methods: Using 33 Box-Behnken designs, about 17 experiments were performed and evaluated for various parameters like physical appearance, pH, in vitro gelling study, in vitro buoyancy study, measurement of viscosity, density measurement, gel strength, raft resilience, drug content, acid neutralization capacity, in vitro dissolution, release kinetics and stability studies. Results: All the formulations exhibited good viscosity, density less than gastric fluid, gelling capacity retained, and buoyant for 12 h. Drug content ranges from 97.98 to 99.34 %, with a long neutralization period. The buoyancy lag time was found to be in the range of 15.34 to 26.12 sec and the % drug release at 12 h was between range from 85.67 to 99.45, with the highest release exhibited by F3. All formulations displayed zero order in vitro drug release>10 h with exceptional buoyancy properties. F3 was the optimized formulation and further subjected to FTIR and DSC study, concluding that compatibility of nizatidine with excipients in the formulation blend. Stability studies show no significant changes. Conclusion: Results indicate that gastric-floating formulations of nizatidine have the prospective for superior gastric residence time and sustained drug release.
