Formulation and evaluation of simvastatin buccal film

Abstract: The aim of this work was to develop and evaluate a buccal film for delivery of simvastatin. Buccal films containing carboxymethyl cellulose (CMC) with and without Polyvinylpyrrolidone (PVP) as polymers were prepared. Tween 80 was included with and without propylene glycol (PG) to modulate the characteristics of the films. The films were prepared by solvent casting with drug concentration being maintained at 2.5 mg/cm 2 . In absence of Tween and PG, the films were opaque due to precipitation of drug. PG reduced… Show more

“…Accordingly, faster swelling of the polymer led to rapid initiation of diffusion in addition to the formation of adhesive bonds. In our work, the swelling index was constant but lower than that reported by El Maghraby and Abdelzaher (2015). These findings indicated that the water absorption power decreased not only as a result of using propylene glycol as a plasticizer (Basu et al, 2010; Anjankumar, 2011) but also due to the presence of polyvinyl alcohol.…”

“…The solubility of urea and propylene glycol might lead to drug diffusion to the dissolution medium. In addition, the role of tween 80, which normally used in pharmaceutical formulation as a release enhancer, should also be considered (Basu et al, 2010; Anjankumar, 2011; El Maghraby and Abdelzaher, 2015). …”

“…Among these, the optimization technique is simply finding the independent variable values that result in minimizing or maximizing the objective function subject to constraints, and the desirability functions are used to solve complicated situations that occurred during optimization techniques. 20,39 Response surface methodology has been successfully utilized in several studies to optimize process and formulation variables and to obtain a product with desired properties. 40 Box-Behnken design is one of the efficient response surface methodology tools used mainly to explore and evaluate the main effect, the interaction and the quadratic terms of formulation, and the process variables on the quality attributes of the formulation.…”
