Formulation Development, Characterization and Antifungal Evaluation of Chitosan NPs for Topical Delivery of Voriconazole In Vitro and Ex Vivo

Abstract: This study aims to develop chitosan-based voriconazole nanoparticles (NPs) using spray-drying technique. The effect of surfactants and polymers on the physicochemical properties, in vitro release, and permeation of NPs was investigated. The prepared NPs containing various surfactants and polymers (e.g., Tween 20 (T20), Tween 80 (T80), sodium lauryl sulfate (SLS), propylene glycol (PG), and Polyethylene glycol-4000 (PEG-4000)) were physiochemically evaluated for size, zeta potential, drug content, percent entra… Show more

“…The amide bond formation between the chitosan and folic acid was evidenced by a shift of the FTIR wavenumber of folic acid from 1760 to 1680 cm −1 . The assignment of FTIR peaks was correlated with earlier studies [ 20 , 21 , 22 ]. The peaks at 2.07 ppm attributed to the acetamino group CH3, and the CH peak appeared at 3.50–3.95 ppm, corresponding to carbons 3, 4, 5, and 6 of the glucosamine rings of CS.…”

“…The tensile strength of LS-TC-MNP was measured by using the method of Khan et al [ 33 ]. The LS-TC-MNP was placed between the jaws of auto tensile tester (Suzhou Tophung Machinery Equipment Co., Ltd., Jiangsu, China) until the LS-TC-MNP split in two parts.…”

“…The percentage elongation of the LS-TC-MNP was measured until it was divided into two parts. An auto tensile testing machine was used to measure the starting and ending length of the LS-TC-MNP and the elongation at breaking point was calculated using Equation (2) [ 33 ]. …”

“…This might be due to increased cross linking of GA. Generally, mobility of the polymer chain is affected by the crosslinking, ensuring the low solubility of the polysaccharide [ 37 ]. GA promotes the degree of crosslinking which in turn results in folding of the polymeric chains and the subsequent attainment of reticulation point, thus affecting the aqueous absorption capacity [ 38 ].…”
