Forward genetic screen for malignant peripheral nerve sheath tumor formation identifies new genes and pathways driving tumorigenesis

Abstract: Malignant peripheral nerve sheath tumors (MPNSTs) are sarcomas of Schwann cell-lineage origin that occur sporadically or in association with the inherited syndrome, Neurofibromatosis Type 1. To identify genetic drivers of MPNST development, we utilized the Sleeping Beauty (SB) transposon-based somatic mutagenesis system in mice with somatic loss of tumor protein p53 (Trp53) function and/or overexpression of epidermal growth factor receptor (EGFR). Common insertion site (CIS) analysis of 269 neurofibromas and 1… Show more

“…In addition, in many cases, only a single transposon insertion was present at a CCG in tumor cells, suggesting that many CCGs are functioning as haploinsufficient TSGs. This is similar to what has been observed in other SB mutagenesis screens performed in solid tumors (18)(19)(20)(21)(22)(23)(24)(25)(26).…”

“…Identification of Trunk Driver Genes. Most CCGs identified by SB mutagenesis are thought to function during late stages of tumor progression (18)(19)(20)(21)(22)(23)(24)(25)(26). To identify the CCGs that function early in mammary tumor development, we selected transposon insertion sites represented by the highest number of sequencing reads (SB insertions in the CCG in ≥3 tumors with ≥9 reads per tumor), arguing that these insertions would be present in the largest number of tumor cells.…”

“…101 Examination of the common integration sites in 269 neurofibromas and 106 MPNSTs identified 695 and 87 genes, respectively, that were recurrently present in these tumors; 37 of these genes were detected in both neurofibromas and MPNSTs. Intriguingly, tumor suppressor genes predominated in these neoplasms, with 67 potential tumor suppressor genes and 20 candidate oncogenes identified.…”

“…High-throughput whole-genome analysis with microarrays has provided the most insight into new therapeutic options by revealing patterns or molecular signatures common in MPNSTs that are not present in benign neurofibromas, that is, copy number variations and gene expression changes (15,16). It has been previously determined that the HGF, MET, and PDGFRA genes are frequently amplified in MPNSTs (15,17).…”
