Cosmetic manufacturers are continuously searching for new active ingredients to satisfy consumer needs. This leads to growing demand of botanical extracts with characterized chemical composition and proven efficacy. The aim of the study was to characterize the chemical composition of birch sap and a complex of seven plant extracts, with brand name Cell Repair Bio-Complex (CRB), and to evaluate their effects on skin cells. HPLC analysis was used to identify dominating compounds. Effect on proliferation of dermal fibroblasts and keratinocytes was assessed using the IncuCyte ZOOM system. Results show that flavonoid and phenolic acid rich CRB possesses antiradical activity and stimulates dermal fibroblast proliferation at low concentration. Birch sap stimulates both dermal fibroblasts and keratinocytes at high concentrations. Stimulatory effect of keratinocytes was observed also for the combination of birch sap and CRB. The findings indicate the potential of CRB to be used as an active ingredient, whereas birch sap used at high concentrations could substitute water base and add additional efficacy.Key words: birch sap, dermal fibroblasts, epidermal keratinocytes, plant extracts, skin antiageing. Amaç: Alchemilla L. cinsine ait türlerin genellikle toprak üstü kısımlarının halk arasında diüretik, astrenjan, antispazmodik, yara iyileştirici, aşırı menstürasyonda kür olarak ve konvulsif hastalıkların tedavisinde kullanılmakta olduğu bilinmektedir. Sahip oldukları zengin polifenol içeriği sebebiyle de antioksidan, antimikrobiyal ve antienflamatuvar etkiler gösterdiği belirlenmiştir. Bu çalışmada ise cinse ait türlerden A. mollis bitkisinin köklerinden hazırlanan ekstrelerin antioksidan ve sitotoksik etkilerinin belirlenmesi için çeşitli aktivite testleri yapılarak, etkinliğinin tespit edilmesi amaçlanmıştır. Yöntem: Bu çalışmada A. mollis türünün köklerinden hazırlanan metanol ekstresi ve bu ekstrenin partisyonu ile elde edilen farklı polariteye sahip alt ekstrelerin toplam fenol ve flavonoit miktar tayini ve antioksidan kapasitelerini belirlemek için DPPH ve ABTS radikalini süpürücü etkileri tespit edilmiştir. Ayrıca ekstrelerin L929 ve MDA-MB-231 hücre hatlarında sitotoksik etkileri değerlendirilmiştir. Bulgular: Yüksek fenolik içeriğine bağlı olarak, bitkinin kök ekstrelerinde yüksek antioksidan kapasite tespit edilmiştir. Bitkinin köklerinden elde edilen ekstrelerin L929 sağlıklı fare fibroblast hücrelerinde ve MDA-MB-231 meme kanseri hücre hattında elde edilen sonuçlara göre; ekstreler belli bir konsantrasyon aralığında sağlıklı hücrelere toksik etki göstermezken meme kanseri hücreleri üzerinde belirli dozlarda toksik etkili bulunmuştur. Sonuç: Sonuç olarak A. mollis bitkisi zengin fenolik bileşik içeriği ile kanserli hücrelerin poliferasyonunu inhibe ederek kanser tedavisinde umut verici terapötik ajanların keşfedilmesinde önemli bir kaynak olabileceği tarafımızca yapılan çalışmalarla tespit edilmiştir

Abstract:The aerial parts of Alchemilla L. species (Rosaceae) are used internally as diuretic, laxative, tonic and externally for wound healing in Turkish folk medicine. Antioxidant effects of the extracts, fractions and isolated compounds from the aerial parts of A. barbatiflora Juz. were investigated with following methods: 1,1-diphenyl-2-picryl-hydrazyl (DPPH), and superoxide radical scavenging (SOD), phosphomolibdenum-reducing antioxidant power (PRAP), ferric-reducing antioxidant power (FRAP) assays. In addition, tyrosinase, α-glucosidase and acetylcholinesterase inhibition activities of samples were analyzed. The methanol extract from the aerial parts of plant was consecutively fractionated into four subextracts; n-hexane, chloroform, and remaining water extracts. Further studies were carried out on the most active water subextract and the fractions obtained from water subextract with column chromatography. Phytochemical studies on active fractions of the water subextract led to the isolation of seven metabolites including catechin (1) and a catechin dimer; procyanidin B3 (2), a flavonol glucuronide; quercetin-3-O-β-D-glucuronic acid (miquelianin) (3) with flavonoid glycosides; quercetin-3-O-β-D-galactoside (hyperoside) (4), quercetin-3-O-β-D-arabinoside (guaiaverin) (5), kaempferol-3-O-β-Dxylopyranoside (6) and kaempferol-3-O-(6″-coumaroyl-β-D-glycoside) (tiliroside) (7). Their structures were elucidated by spectral techniques (1D and 2D NMR). The experimental data verified that procyanidin B3 displayed remarkable enzyme inhibitory activity among the whole isolated compounds.
