Frequencies of Memory T Cells Specific for Varicella‐Zoster Virus, Herpes Simplex Virus, and Cytomegalovirus by Intracellular Detection of Cytokine Expression

Abstract: Memory T cells specific for varicella-zoster virus (VZV), herpes simplex virus (HSV), and human cytomegalovirus (HCMV) were compared in immune adults by intracellular cytokine (ICC) detection. The mean percentages of CD4+ T cells were 0.11% for VZV and 0.22% for HSV by interferon (IFN)-gamma production; the frequency for HCMV was significantly higher at 1.21%. Percentages of VZV-, HSV-, and HCMV-specific CD4+ T cells were similar by use of tumor necrosis factor (TNF)-alpha. HCMV-stimulated CD8+ T cells produce… Show more

“…Prior to treatment, VZVspecific T cell response was mainly detected in the CD4 ϩ T cell subpopulation as only one patient had a CD8 ϩ T cell response. A similarly low frequency of VZV-specific CD8 reactivity has been observed in healthy adults, 29 indicating that VZV-induced CD4 ϩ memory T cell response is prevalent and more persistent than the CD8 memory response.…”

“…In healthy adults a similarly high level has been explained by a requirement of a more active immune surveillance to restrict replication of persistent CMV in cells of the monocyte/macrophage lineage. 29 After transplantation CMV-specific T cell recovered within months to the same or an even higher level without any of the patients having clinical symptoms or signs compatible with CMV disease. A similar association has been observed after bone marrow transplantation.…”

“…The mean frequency of whole-HSV Agspecific memory CD4 T lymphocytes is reported to be ∼0.2% of total circulating CD4 T lymphocytes (78). Less than 5% of NK cells (probably mostly the CD56 bright CD16 2 subset) were involved in ISs, also correlating with the proportion of NK cells expressing HLA-DR (data not shown).…”

“…In addition, frequencies of VZV‐specific memory T cells are reportedly low and amount to only half of HSV‐ and to less than a tenth of CMV‐specific T‐cell frequencies in immune adults (Asanuma et al., 2000; Tyler, 2015). Together, this may add to a disrupted defense against VZV reactivation from latency.…”
