Frequency of germline mutations in cancer susceptibility genes in malignant mesothelioma.

Abstract: The aim of the current study was to determine the prevalence and clinical predictors of germline cancer susceptibility mutations in patients with malignant mesothelioma (MM). MethodsWe performed targeted capture and next-generation sequencing of 85 cancer susceptibility genes on germline DNA from 198 patients with pleural, peritoneal, and tunica vaginalis MM. ResultsTwenty-four germline mutations were identified in 13 genes in 23 (12%) of 198 patients. BAP1 mutations were the most common (n = 6; 25%). The rema… Show more

“…This study is amongst the first to elaborate on somatic BRCA mutations in malignant mesothelioma. The distribution of somatic BRCA mutations seen in this study mirrors the findings in germline BRCA with a higher frequency of BRCA2 mutations as compared to BRCA1 mutations [ 10 , 33 , 37 ]. The BRCA2 somatic alterations were limited to single nucleotide polymorphism in all cases similar to pre viously reported germline findings [ 10 ].…”

“…Panou et al, in their targeted capture and next generation sequencing study of 85 cancer susceptibility genes on germline DNA from 198 malignant mesothelioma patients, found BRCA1 germline mutations in 0.0051 proportion of mesothelioma patients included in their study compared to 0.002 estimate in the non-cancer population (p>0.05). In the same study, BRCA2 germline mutations were seen in 0.0152 proportion of mesothelioma patients as compared to the 0.003 estimate in non-cancer population (p=0.03) [ 10 ]. This translates to a prevalence of 8.1% for BRCA2 variants in malignant mesothelioma compared to 2.1% for BRCA1 in this US based study.…”

“…Germline mutations that predispose to MPM were first identified in BAP1, but two recent studies also highlighted germline mutations in several other genes that are less common than in BAP1. They are mainly involved in cell-cycle, chromatin regulation and DNA repair (20)(21)(22)(23)(24). Up to 7% of MPM patients may have germline mutations, but experimental validations are needed to confirm that some of these genes are MPM susceptibility genes (like BAP1).…”

“…Although asbestos use is banned in many countries, asbestos-insulated buildings are present throughout the world and some countries are still manufacturing and using large quantities of asbestos (2). Germline mutations in BAP1 and in other cancer susceptibility genes such as PALB2, BRCA2, CHEK2 and MLH1 have been identified in about 10-15% of patients with MPM (3)(4)(5)(6)(7).…”
