“…Closely related proteins include sequences from the wolf spider Pardosa pseudoannulata and the tick Rhipicephalus sanguineus that share 67% and 63% similarity with CsnAChα ( Fig 5 ). Both of these sequences have been expressed in Xenopus oocytes, but similarly to most invertebrate nACh subunits, they failed to function as homomultimers [ 8 , 70 ]. When co-expressed with nAChβ subunits, they formed functional ACh and nicotine sensitive receptors.…”

“…Mammalian Cys‐loop receptors include anion‐selective γ‐aminobutyric acid type A (GABA A ) and glycine receptors, and cation‐selective nicotinic acetylcholine (nACh) and 5‐HT 3 receptors (Miller and Smart, ). Invertebrates have several types of GABA A and nACh receptors as well as glutamate‐ (GluCl), histamine‐ (HisCl), and pH‐ (pHCl) gated Cl channels (Hosie et al, ; Ozoe et al, ; Lees et al, ). Invertebrate Cys‐loop receptors are prime targets for widely used control agents against agricultural pests and disease vectors (Tomizawa and Casida, ; Wolstenholme, ).…”

“…Using R. sanguineus nAChR α1 subunit, it was found that its expression into X. laevis oocytes failed to function as a homomer. When co-expressed with chicken β2 subunit, the chimeric α1β2 nAChR evoked inward currents with a slow activation and deactivation kinetics, in response to ACh and nicotine ( Lees et al, 2014 ). We however argued that the functional properties of expressed receptors could be influenced by the host heterologous cell expression system, in terms of activation and deactivation currents induced by agonists.…”

“…Transcriptomic analyses on the tick Rhipicephalus sanguineus and Dermacentor variabilis synganglia have shown that they expressed cDNAs encoding for nAChR subunits ( Lees et al, 2010 ; Bissinger et al, 2011 ). Recently, Lees et al, demonstrated that R. sanguineus α1 subunit co-expressed with the chicken β2 subunit, in the Xenopus oocytes , evoked acetylcholine (ACh) and nicotine currents, but the expressed hybrid receptor was insensitive to IMI ( Lees et al, 2014 ). We propose that different nAChR subtypes exist in the tick nervous system and account for the binding of neonicotinoid insecticides.…”
