Functional Core/Shell Drug Nanoparticles for Highly Effective Synergistic Cancer Therapy

Abstract: Gold (Au)-nanoshelled 10-hydroxycamptothecin nanoparticles (HCPT NPs) are developed with combination of photothermal therapy and chemotherapy for highly effective cancer therapy. The strong near-infrared (NIR) absorbance from Au nanoshells endows the nanocomposites photothermal effects and on-demand drug release. Notably, the drug-loading content reaches up to 63.7 wt%, which is much higher than that in the previously reported nanovehicles systems. Both in vitro and in vivo studies indicate that the combined l… Show more

“…The tumor sizes and body weight of mice were measured every day lasting for 15 d after treatment. The tumor volume and relative tumor volume were calculated by the following equations . All measurements were performed in triplicateTumor volume = L×W22where L stands for the longest diameter and W is the shortest diameterRelative tumor volume =normalVVwhere V is the tumor volume after treatment and V ′ is the initial tumor volume.…”

“…Upon laser irradiation, the photothermal heating of the gold nanoshells induced the micelles to collapse and resulted in doxorubicin release [63]. This photothermal approach can be used to induce deformation and rupture of gold nanoshells, where the therapeutic payload has been encapsulated, thus triggering its controlled release [84,[86][87][88]. A mechanism describing how drugs are released from these AuNMs when combined with laser-induced thermal therapy has also been proposed [89].…”

“…[8][9][10] Photothermal therapy (PTT) is an approach to employ photothermal agents with a strong absorbance in the near-infrared (NIR) region to produce heat to kill target cells with minimal side effects. During last decades, a wide range of PTT agents such as gold nanomaterials (e.g., gold nanorods, nanocages, and nanoshells), [11][12][13][14][15] carbon nanomaterials (e.g., carbon nanotubes, and graphene) [16][17][18][19][20] and many other materials (e.g., polydopamine, poly(4-styrenesulfonate), and indocyanine green (ICG)) [21][22] have been widely explored. Chemotherapy is to use drugs to destroy cancer cells.…”
