Functional Proteomics Reveals the Biochemical Niche of C. elegans DCR-1 in Multiple Small-RNA-Mediated Pathways

Abstract: In plants, animals, and fungi, members of the Dicer family of RNase III-related enzymes process double-stranded RNA (dsRNA) to initiate small-RNA-mediated gene-silencing mechanisms. To learn how C. elegans Dicer, DCR-1, functions in multiple distinct silencing mechanisms, we used a mass-spectrometry-based proteomics approach to identify DCR-1-interacting proteins. We then generated and characterized deletion alleles for the corresponding genes. The interactors are required for production of three species of sm… Show more

“…There is an increasing number of reports that Argonautes associates with complexes that contain Dicer, the key enzyme in siRNA and miRNA maturation (Tabara et al, 2002;Sasaki et al, 2003;Okamura et al, 2004;Gregory et al, 2005;Maniataki and Mourelatos, 2005;Meister et al, 2005;Duchaine et al, 2006). The co-association of RISC and the miRNA processing is not surprising.…”

“…The DexDh exonuclease eri-1 is an endogenous inhibitor of RNAi, whereas lin-15b is implicated in chromatin remodeling and also enhances the response to dsRNAs. eri-3 acts in a complex with eri-1 and like mutations in eri-1; eri-3 mutations enhance the response to dsRNAs that are not effective in wild type (Duchaine et al 2006).…”

“…Because Eri1 encodes a DEDDh-type exonuclease with RNase activity toward the 3′ ends of RNA in vitro 1,[4][5][6] , it was proposed that Eri1 counteracts the RNAi pathway by degrading siRNAs 7 . Alternatively, Eri1 may inhibit RNAi indirectly by sequestering factors involved in siRNA processing into a different protein complex [8][9][10] .…”
