Functionalized magnetic nanobeads for SERS-based detection of Staphylococcus aureus

Abstract: We introduced a Raman detection technique based on a combination of functionalised magnetic beads and surface-enhanced Raman scattering (SERS) tags to develop a rapid and sensitive strategy for the detection of Staphylococcus aureus.

“…Polyethylene glycol (PEG) and bovine serum albumin (BSA) dual-mediated teicoplanin-functionalized magnetic beads (TEI-BPBs) were used for the isolation of the target bacteria. TEI can recognize and capture Gram-positive bacteria, such as S. aureus , by binding to the D -Ala– D -Ala peptide fraction; however, it lacks some specificity in recognizing the bacterial cells [ 234 ]. Therefore, SERS tags were used to immobilize antibodies on gold surfaces via bifunctional linker proteins to ensure specific recognition of S. aureus .…”

“…Following separation, the bacteria undergo Raman spectroscopy to acquire their unique spectral signatures. A novel method for detecting S. aureus, a common foodborne pathogen, was developed by Ji et al [234], using functionalized magnetic beads and surface-enhanced Raman scattering (SERS) tags. Polyethylene glycol (PEG) and bovine serum albumin (BSA) dualmediated teicoplanin-functionalized magnetic beads (TEI-BPBs) were used for the isolation of the target bacteria.…”
