“…In the context of aging studies, it is crucial to ensure that the system does not allow any gene expression in absence of inducer during pre-adult stages. As a stringent test of leakiness (Seroude, 2002), each Gene-Switch driver was combined with a UAS-Grim reporter (encoding a strong pro-apoptotic factor) and embryonic, larval and pupal lethality was scored in absence of inducer and compared to the negative control carrying the reporter alone. Lethality was also measured in presence of 50 μg mL -1 of RU486 to confirm that Grim expression would indeed cause developmental lethality.…”
