Gambierone, a Ladder-Shaped Polyether from the Dinoflagellate Gambierdiscus belizeanus

Abstract: A new natural product named gambierone (1) was isolated from the cultured dinoflagellate Gambierdiscus belizeanus. The structure of this compound features an unprecedented polyether skeleton and an unusual right-hand side chain. Its relative configuration was fully determined by interpretation of ROESY experiment and comparison between experimental and theoretical NMR data. Although the succession of cycles has no chemical similarity with ciguatoxins, 1 has a molecular formula and biological activity similar t… Show more

“…14 This approach based on theoretical NMR calculations has been proven to be a very powerful tool in natural products structure elucidation. 9,15 Taking both 1 H and 13 C chemical shifts calculations for the DP4 probability analysis ( Figure S40), the relative configuration at C-9a was defined to be R.…”

“…For that reason, the authors carried out NMR calculations of the eight resulting candidates at the B3LYP/6‐31G(d)//B3LYP/STO‐3G level. The DP4 probability computed from 13 C data strongly supported the 2 R *,4 R *,38 S * stereochemistry present in 28 (91.2 %) that was finally proposed for this complex natural product …”

“…Gambierdiscus species also synthesize other toxins, including gambieric acids [81], gambierol [82], gambierone [83] and maitotoxins [84,85]. Maitotoxin (MTX-1) itself is a water-soluble cyclic polyether ladder marine toxin that was first described in the mid-1970s (Table 1).…”
