Gene cluster lock after pheromone receptor gene choice

Abstract: In mammals, perception of pheromones is based on the expression in each vomeronasal sensory neuron of a limited set of receptor genes, chosen among a large repertoire. Here, we report an extremely tight control of the monogenic and monoallelic transcription of the V1rb2 receptor gene. Combining genetic and electrophysiological approaches, we show that the transcription of a nonfunctional V1r allele leads to the coexpression of another, functional V1r gene. The choice of this coexpressed gene surprisingly inclu… Show more

“…It is known that a substantial number of pseudogenes are present in the mammalian OR gene families (Ϸ30% of total OR genes in the mouse and Ϸ60% in human are nonfunctional). If an activated LCR has selected a pseudogene and has been trapped by its promoter, other LCRs must undergo a similar process to ensure the activation of an intact OR gene (32). Pseudogene may help slow down the process of OR gene activation and further reduce the frequency of the simultaneous activation of two different OR genes.…”

“…In situ hybridizations were performed as described previously (Roppolo et al, 2007). Briefly, tissues were embedded in OCT and 14 m cryostat sections were mounted on Superfrost Plus slides.…”

“…G protein-coupled vomeronasal chemoreceptor proteins are encoded by 3 unrelated multigene families-the V1r (Dulac and Axel 1995), V2r (Herrada and Dulac 1997;Matsunami and Buck 1997;Ryba and Tirindelli 1997), and Fpr-rs (Liberles et al 2009;Rivière et al 2009) genes. Monogenic receptor expression and type-specific coexpression of G protein α-subunits (G αi2 /G αo ) confer a distinct receptor "identity" to each VSN (Roppolo et al 2007) and result in topographical segregation into at least 2 VSN subpopulations.…”
