Generating an oilseed rape mutant with non-abscising floral organs using CRISPR/Cas9 technology

Abstract: Oilseed rape plants with abscission-defective floral organs acquired through genome editing show less susceptibility to Sclerotinia sclerotiorum infection and longer flowering-period for flower tourism.

“…For example, knockdown of the gibberellin synthesis gene GA REQUIRING 4 ( BnGA4s ) in B. napus not only affects the development of the cotyledonary septum and improves resistance to silique shattering, but also leads to shorter plant height (Stephenson et al, 2019). Knockdown of INFLORESCENCE DEFICIENT IN ABSCISSION ( BnIDA) causes dehiscence of the angiosperms along with abscission of the floral organs and reduced resistance to Dictyostelium nucleatum (Geng et al, 2022; Wu et al, 2022). In the future, strengthening the understanding of traits related to suitable machine harvesting can be beneficial to promote the efficiency of mechanized harvesting of B. napus , and increase yield.…”

“…Gene redundancy in polyploid species could also mask the mutant genotypes even when the gene knockout occurred in the target site. 15 Double mutants were developed through crossing lines containing either of single mutant gene bnc06.ida and bna07.ida involved in floral abscission of rapeseed. 15 DNA markers were used to discriminate between heterozygous and homozygous genotypes and finally doubled mutants were recovered in the F 2 population through speed breeding within six months.…”
