Generation, annotation and analysis of ESTs from Trichoderma harzianum CECT 2413

Abstract: Background: The filamentous fungus Trichoderma harzianum is used as biological control agent of several plant-pathogenic fungi. In order to study the genome of this fungus, a functional genomics project called "TrichoEST" was developed to give insights into genes involved in biological control activities using an approach based on the generation of expressed sequence tags (ESTs).

“…Starting in 2002, the TrichoEST functional genomics project sequenced more than 25,000 ESTs and described 13,814 unique transcripts from eight different species representing the biodiversity of this genus: T. harzianum, T. virens, T. atroviride, T. asperellum, T. viride, T. longibrachiatum, T. stromaticum, and T. aggressivum (99). The ESTs were from 28 cDNA libraries obtained under a wide range of growth conditions, including biocontrol interactions and nutrient stress (140). A subset of 8710 ESTs, from eight T. harzianum CECT2413 (T. harzianum T34) cDNA libraries, revealed 3478 unique sequences.…”

“…Trichoderma spp. can also exert marked antimicrobial activity (Vizcaíno et al ., ) as a result of the production of a huge diversity of secondary metabolites (Cardoza et al ., ; Reino et al ., ), including, for example, pyrones [6‐pentyl‐2H‐pyran‐2‐one (6‐PP) derivatives] (Keszler et al ., ; Rubio et al ., ), butenolides (Cai et al ., ), peptaibols (Vizcaíno et al ., 2005b; 2006), and trichothecenes (terpenes) (Cardoza et al ., ; Malmierca et al ., 2012; 2013).…”

“…Two bands of 21 and 32 kDa (indicated by arrowheads) were observed consistently in all the strips. M molecular weight markers, C crude extract of T. harzianum on 12% gel consistent with the outcomes of a genome analysis study of T. harzianum strain CECT 2413 (Vizcaíno et al 2006) where a total of 3,478 unique sequences were identified with one having sequence similarity with Alt a 5, an allergen reported from Alternaria alternata (I.U.I.S. 2007).…”
