Genetic characterization and phylogenetic analysis of porcine circovirus type 2 (PCV2) in Serbia

Abstract: Porcine circovirus type 2 (PCV2) is the main causative agent of postweaning multisystemic wasting syndrome (PMWS). To characterize and determine the genetic diversity of PCV2 in the porcine population of Serbia, nucleotide and deduced amino acid sequences of the open reading frame 2 (ORF2) of PCV2 collected from the tissues of pigs that either had died as a result of PMWS or did not exhibit disease symptoms were analyzed. Sequencing and phylogenetic analysis showed considerable diversity among PCV2 ORF2 sequen… Show more

“…No isolates from group PCV2a were found in 2012 indicating that this viral group has spontaneously decreased in its spread during the last decade or has been eliminated from the tested farms as a result of vaccination. Similar findings were reported and recognised in countries neighbouring Croatia, such as Slovenia [25], Hungary [26], Austria [27], Serbia [28,29], and Italy [30]. Vaccination against PCV2 infection in Croatia was introduced rather late, in 2010.…”

“…Each genetic cluster was found at more than one site and indicates the co‐circulation of viruses among pig farms. In recent years, PCV2 viruses bearing a lysine extension at the C‐terminus of the capsid protein have been reported in several countries, including Germany (Knell et al., ), Indonesia (Manokaran et al., ), China (Wang et al., ; Guo et al., ) and Serbia (Savic et al., ). In addition, the PCV2b genotype with the capsid protein mutation has been confirmed to display increased virulence in vivo compared to classical PCV2a and PCV2b genotypes (Guo et al., ).…”

“…The lysed samples were processed with a DNA extraction kit (QIA extraction DNA kit, Qiagen) according to the manufacturer's instructions. All samples were tested by PCR using HotStarTaq Master Mix Kit (Qiagen), and primers described for DNA detection of PCV2 [11], Mhp [12], Pm [13], App [14], Hps [15], Ssuis [16] and Apyo [17]. After suspension, the samples were processed with a RNA extraction kit (Qiagen -Rneasy MiniKit) according to th e manufacturer's instructions.…”
