Genetic Identity between Bhadawari and Murrah Breeds of Indian Buffaloes (Bubalus bubalis) Using RAPD-PCR

Abstract: Randomly Amplified Polymorphic DNA-Polymerase Chain Reaction (RAPD-PCR) analysis was carried out with a battery of 11 random decamer primers to study band frequency (BF), genetic identity index (I) and mean average percentage difference (MAPD) between Bhadawari and Murrah breeds of buffalo. The primers OPA04 and BG15 resolved a band of 460 bp, which was present only in animals of Bhadawari breed. Whereas, the primers OPA14, BG27 and BG28 produced Murrah specific fragments of sizes 730 bp and 1,230 bp, respecti… Show more

“…The number of fragments amplified in four breeds ranged from 2 to 10 fragments per primer. The number of DNA fragments amplified with each primer found in the present study is in agreement with several investigators (Chen et al, 2001;Rehman, 2002;Appa Rao et al, 1996;Ali, 2003;Kumar et al, 2003;Saifi et al, 2004;Mahfouz, 2008). Kumar et al (2003) used six oligonucleotide primers for RAPD fingerprinting in order to estimate genetic relationship among four different breeds of sheep.…”

“…The DNA-RAPD assay which is short oligonucleotide primers of arbitrary sequence to amplify genomic DNA by PCR enables the approach for identifying genetic markers (Eman, et al, 2008). RAPD was utilized in breeds genetic studies in most species, including farm animals like cattle , buffalo (Saifi, et al, 2004), goat (Chen, et al, 2001), chicken (Okumus and Kaya, 2005) and sheep (Okumus and Mercan, 2007). The investigation of genetic diversity and similarity among and within breeds is necessary be providing useful genetic information essential for developing affective management plans for the conservation and improvement of these genetic resources.…”
