Genetic Linkage Mapping and Analysis of Muscle Fiber-Related QTLs in Common Carp (Cyprinus carpio L.)

Abstract: A genetic linkage map of common carp (Cyprinus carpio L.) was constructed using Type I and Type II microsatellite markers and a pseudo-testcross mapping strategy. The microsatellite markers were isolated from microsatellite-enriched genomic libraries and tested for their segregation in a full-sib mapping panel containing 92 individuals. A total of 161 microsatellite loci were mapped into 54 linkage groups. The total lengths of the female, male and consensus maps were 2,000, 946, and 1,852 cM, with an average m… Show more

“…Significantly, the unique dual reproduction mode, as commented by Avise (2008), was the first documented instance in vertebrates [19], and the discovery firstly explained reproduction mechanism of high genetic diversity and an evolutionarily long-lived timescale of unisexual species [18]. In common carp (Cyprinus carpio), along with advances of polymorphic markers and genome sequencing, various density linkage maps were constructed [20][21][22][23], and used to locate cold tolerance [20] and muscle fiber-related [22] QTLs. A physical map was constructed from a BAC library containing a total of 92160 BAC clones with an average insert size of 141 kb [24,25].…”

“…In addition, it is also considered an alternative vertebrate fish model to zebrafish (Danio rerio). A variety of C. carpio genome resources have been developed over the past decade, including a large number of genetic markers 8,9 , genetic maps [10][11][12][13] , a BAC-based physical map 14,15 , a large number of ESTs [16][17][18] and cDNA microarrays 19 . Recently, a comparative exomic study of C. carpio and D. rerio has been reported, providing additional genome resourcing data for the research community 20 .…”

“…Many quantitative trait loci (QTLs) have been identified for various economic traits, such as cold-tolerance [6], growth-related traits [12,13], muscle quality [14], feed conversion efficiency [15], and amino acid content [16] in common carp. However, all of these QTL studies were carried out in populations of progeny derived from single crosses.…”
