Genetic Polymorphisms in Folate Metabolism and the Risk of Stomach Cancer

Abstract: Folate deficiency has been implicated in the etiology of stomach cancer through abnormal DNA methylation and disrupted DNA synthesis and repair. Enzyme-coding genes involved in folate metabolism are often polymorphic. In a population-based study of 305 cases and 427 controls in Warsaw, Poland, we evaluated the risk of stomach cancer in relation to polymorphisms in folate-metabolizing genes, including MTHFR (Ex5+79C>T and Ex8À62A>C), MTR (Ex26À20A>G), and MTRR (Ex2À64A>G, Ex5+123C>T, Ex15+572C>T, Ex15À405A>T, E… Show more

“…Studies investigating the MTRR A66G (rs1801394) polymorphism with breast cancer, gastric cancer, head and neck squamous cell carcinomas, and multiple myeloma have yielded inconsistent results [27][28][29][30][31]. In the present study, no clear evidence of a link between the MTRR A66G polymorphism and cervical carcinogenesis risk was found.…”

“…Several studies conducted in Chinese, Caucasian, and Mexican populations have shown that the MTHFR C677T polymorphism does not influence the risk of gastric cancer (Weng et al, 2006;Götze et al, 2007;Zhang et al, 2007). However, several studies conducted in Korean, Chinese, and Caucasian populations indicated the MTHFR 677TT genotype has a protective effect on the risk of gastric cancer.…”

“…Some studies have shown associations with colon (14), lung (15) and breast cancer (16), and relationships with tumor aggressiveness and variable responses to esophageal cancer treatment (17). Other studies did not confirm associations between the polymorphism and different types of cancer, including colorectal (18), lung (19), bladder (20), and stomach cancer (21).…”
