Genetic polymorphisms in human SULT1A1 and UGT1A1 genes associate with breast tumor characteristics: a case-series study

Abstract: IntroductionEstrogens are important in breast cancer development. SULT1A1 and UGT1A1 catalyze estrogen metabolism and are polymorphic. The SULT1A1*2 protein exhibits low activity, and a TA repeat within the UGT1A1 promoter alters the level of expression of the protein. We hypothesized that the SULT1A1*2 allozyme has decreased capacity to sulfate estrogens, that the SULT1A1*2 allele conferred increased capacity of cells to proliferate in response to estrogens, and that individuals with the variant SULT1A1 and U… Show more

“…Additional assay validations were undertaken using direct sequencing (CYP1A1*2C), pyrosequencing (SULT1E1 À64G>A), and RFLP (COMT, CYP1B1*4, CYP3A4*1B). SULT1A1 genotypes were determined as previously reported (5,16). The control frequency distributions for these variants by race has also been previously reported (5).…”

“…Recently, mutations in the SULT1E1 gene have also been reported; however, the mutation frequencies in the population studied were very low (Adjei et al, 2003). The possible association between SULT1A1 pharmacogenetics (SULT1A1 normal-activity *1 and low-activity *2 alleles) and cancer has been evaluated in several studies (Seth et al, 2000;Bamber et al, 2001;Zheng et al, 2001;Wang et al, 2002;Tang et al, 2003;Wu et al, 2003;Choi et al, 2005;Nowell et al, 2005;Shatalova et al, 2005). These studies provide epidemiological evidence for associations among diet, genetics, and cancer risk.…”

“…One study was excluded as all the required information could not be obtained (Peng et al, 2003). Genotyping studies on only cancer cases or exclusively on healthy subjects were excluded as comparison of cancer patients with matched controls was a prerequisite for studying association of a particular genotype with cancer risk (Nowell et al, 2002b(Nowell et al, , 2005Magagnotti et al, 2003;Sparks et al, 2004;Shatalova et al, 2005;Grabinski et al, 2006).…”
