Genome editing reveals dmrt1 as an essential male sex-determining gene in Chinese tongue sole (Cynoglossus semilaevis)

Abstract: Chinese tongue sole is a marine fish with ZW sex determination. Genome sequencing suggested that the Z-linked dmrt1 is a putative male determination gene, but direct genetic evidence is still lacking. Here we show that TALEN of dmrt1 efficiently induced mutations of this gene. The ZZ dmrt1 mutant fish developed ovary-like testis, and the spermatogenesis was disrupted. The female-related genes foxl2 and cyp19a1a were significantly increased in the gonad of the ZZ dmrt1 mutant. Conversely, the male-related genes… Show more

“…Such technologies as zinc-finger nucleases, transcription activator-like effector nucleases and clustered, regularly interspaced, short palindromic repeats (CRISPR)-CRISPR-associated (Cas) systems (CRISPR/Cas9) have emerged as new tools for potential improvement of aquacultured fish (Cui et al, 2017;Li and Wang, 2017). The CRISPR/Cas9 gene editing technology has been used to generate lines of site directed mutated cell lines of Chinook salmon O. tshawytscha (Dehler et al, 2016) and can be used for determination of SNP function and QTL assessment (Macqueen et al, 2017).…”

“…semilaevis [2]. In the gonads of DMRT1 -knock out fish, no secondary spermatocytes, spermatids and sperms have been detected, and only spermatogonia and primary spermatocytes were found [5]. In addition, ZZZ triploid males cannot produce normal sperm due to the inhibition of testis development [4].…”

“…Triploid males were produced using hydraulic pressure shock, and triploidy was validated by karyotyping and flow cytometry analysis [3]. DMRT1 -knock out fish was also produced in our laboratory by microinjecting the TALEN mRNAs targeted towards dmrt1 into sole embryos (wild-type) [5]. Total RNA was extracted with a TRIzol isolation kit (Qiagen, Dusseldorf, Germany).…”
