Genome-wide transcriptome analysis reveals that a pleiotropic antibiotic regulator, AfsS, modulates nutritional stress response in Streptomyces coelicolor A3(2)

Abstract: Background: A small "sigma-like" protein, AfsS, pleiotropically regulates antibiotic biosynthesis in Streptomyces coelicolor. Overexpression of afsS in S. coelicolor and certain related species causes antibiotic stimulatory effects in the host organism. Although recent studies have uncovered some of the upstream events activating this gene, the mechanisms through which this signal is relayed downstream leading to the eventual induction of antibiotic pathways remain unclear.

“…AfsS may provide some speciesspecific beneficial modulation of AfsR function by virtue of its (probably mostly indirect) influences on the transcription of various stress response and stationary-phase genes (136). More than 117 genes were up-or downregulated in an afsS disruption mutant (136). Disruption of afsS also causes diminished actinorhodin production during phosphate starvation, an observation underpinned by the finding that AfsR and PhoP bind to overlapping sequences in the promoter region of afsS (82).…”

“…AfsS (also called AfsR2) has been studied as an apparent positive regulator of antibiotic production since its dis- covery (135). It has some homology with the conserved linker domain 3 of sigma factors but is unlikely to function as a sigma factor on its own (136). A suggestion that AfsS might act cooperatively with sigma factors of the ECF subclass, which lack domain 3 (136), seems unlikely given that ECF sigma factors are ubiquitous while AfsS-like proteins are found in only some (not all) streptomycetes.…”

“…Examples of these regulators are AfsK, AfsR, and AfsS. These proteins have been shown to influence secondary metabolism and morphogenesis in several species of Streptomyces (8,17,29,30,40,42).…”
