Germ cell differentiation-dependent and stage-specific expression of LANCL1 in rodent testis

Abstract: LANCL1 (LanC-like protein 1) is related to the bacterial LanC (lanthionine synthetase C) family, which is involved in the biosynthesis of antimicrobial peptides. Highest expression levels of LANCL1 are found in testes and brain, two organs that exist behind blood-tissue barriers. In the mouse, the establishment of an impermeable blood-testis barrier occurs between day 10-16 post natal (pn). Differential display analysis showed that the expression level of LANCL1 mRNA in mouse testes was very low until day 16 p… Show more

“…Aliquots of 200 ng were used for in vitro transcription labelling of the RNA probes using MEGAscript-T3 (sense) or MEGAscript-T7 (antisense) following the manufacturer's protocol (Ambion, Applied Biosystems). ISH was performed as described previously (Nielsen et al 2003, Novotny et al 2007b.…”

“…ISH was performed essentially as described previously [20]. The ISH procedure in brief: deparafinised sections (4 m) were re-fixed in 4% parformadehyde (PFA), treated with proteinase K Germany) followed by development with NBT BCIP (262.5 g/mL p-nitro-blue tetrazolium chloride; 225 g/mL 5-bromo-4-chloro-3-indolyl phosphate dipotassium salt dissolved in 100% and 70% dimethyl formamide, respectively) with levamisol (1.25 × 10 −6 M).…”

“…92128, and PC5/6 sense probe. In situ hybridization was performed as described previously (31). In brief, mouse jejunal mucosa was fixed in 4% paraformaldehyde; 8-m sections were placed on SuperFrostRPlus slides (Menzel GmbH, Braunschweig Germany), deparaffinized, and paraformaldehyde (4%) fixed, and treated with proteinase K (P-2308; Sigma, St. Louis MO); postfixed (4% paraformaldehyde); prehybridized for 1 h at 50 C; and incubated overnight with the labeled PC5/6 probe (50 C) specific for nucleotide.…”
