Ghrelin accelerates wound healing through GHS-R1a-mediated MAPK-NF-κB/GR signaling pathways in combined radiation and burn injury in rats

Abstract: The therapeutic effect of ghrelin on wound healing was assessed using a rat model of combined radiation and burn injury (CRBI). Rat ghrelin, anti-rat tumor necrosis factor (TNF) α polyclonal antibody (PcAb), or selective antagonists of p38 mitogen-activated protein kinase (MAPK), c-Jun N-terminal kinase (JNK), and growth hormone secretagogue receptor (GHS-R) 1a (SB203580, SP600125, and [D-Lys3]-GHRP-6, respectively), were administered for seven consecutive days. Levels of various signaling molecules were asses… Show more

“…These increases were suppressed by NBD in a concentration-dependent manner, demonstrating that NBD has significant anti-inflammatory properties, which may explain how NBD can inhibit cytokine production by inhibiting the NF- κ B signaling pathway. Previous studies showed that NF- κ B signaling pathway overactivation is closely related to ALI development [31, 32]. NF- κ B signaling is upregulated to induce the transcription of some inflammatory factors in LPS-induced ALI [12, 33].…”

“…Dixit et al showed the inhibitory function of ghrelin toward inhibitor of kappa B (IκB) and intensification of activity of some cytokines as well as secretion of IL‐17 by T lymphocytes. In a study by Liu et al, it was also found, using an animal model, that administration of ghrelin to rats subjected to ionizing radiation decreased the concentration of TNF‐α, IL‐1β, and IL‐6. The authors demonstrated that application of ghrelin 4 days after irradiation reduced the serum level of TNF‐α by 52% …”
