Glucocorticoids and non-alcoholic fatty liver disease

Abstract: Non-alcoholic fatty liver disease (NAFLD) is the hepatic manifestation of the global obesity and metabolic disease epidemic and is rapidly becoming the leading cause of liver cirrhosis and indication for liver transplantation worldwide. The hallmark pathological finding in NAFLD is excess lipid accumulation within hepatocytes, but it is a spectrum of disease ranging from benign hepatic steatosis to steatohepatitis through to fibrosis, cirrhosis and risk of hepatocellular carcinoma. The exact pathophysiology re… Show more

“…Considering that chronically elevated glucocorticoid levels lead to non-alcoholic fatty liver2627, whereas the expression of Angptl8 is inhibited by glucocorticoid signal, we wondered whether Angptl8 plays a role in this regard. To answer this question, we overexpressed GFP, Angptl8 or Δ25- Angptl8 in the liver by tail-vein delivery of corresponding adenoviruses on day 1, and then injected these mice with either DEX or PBS once-daily for three days from day 4 to day 6.…”

“…Over the past decade, NAFLD has become one of the most prevalent causes of chronic liver disease [40]. Accumulating evidence has shown that psychological stress may correlate with the increasing incidence of metabolic syndrome, inflammation, and NAFLD [41, 42].…”

“…Estimates suggest approximately 3.4 million deaths/year from obesity-associated diseases such as cardiovascular disease, type-2 diabetes (T2D), and certain types of cancer [1], [2], [3], [4]. NAFLD has been recognized as one of the leading causes of liver cirrhosis today [5], [6]. It is present in up to 30% of Americans and is involved in the development of insulin resistance (IR) and T2D [5].…”
