Glucocorticoids drive human CD8+ T cell differentiation towards a phenotype with high IL-10 and reduced IL-4, IL-5 and IL-13 production

Abstract: Glucocorticoids are highly effective in the treatment of allergy and asthma and inhibit the synthesis of IL‐4, IL‐5 and IL‐13 by disease‐promoting CD4+ Th2 cells. CD8+ T cells also synthesize these cytokines, and the aim of this study was to investigate how glucocorticoids effect cytokine production by these cells. When CD8+ T cells are stimulated with anti‐CD3 and IL‐2 plus IL‐4 or dexamethasone, production of the anti‐inflammatory cytokine IL‐10 is low in both primary and secondary cultures restimulated with… Show more

“…Associations between inflammatory markers, the immune system, and depression have also been described (Miller et al 2009;Miller 2010). In animals, the elevation of APPs by transportation and weaning stress has been reported (Richards et al 2000;Arthington et al 2003). Weaning stress may induce an APR through increased cortisol production and modulation of inflammatory cytokines (Kim et al 2011).…”

“…However, not all cytokines are suppressed by glucocorticoids. IL-10 secretion is increased by glucocorticoids (Blotta et al, 1997;Richards et al, 2001), whereas others (i.e. IL-6) act synergistically with glucocorticoids (Wiegers et al, 2005).…”

“…Another possibility is that GCR action leads to enhanced serine-phosphorylation As discussed above GC can also induce IL-10 in monocytes (Mozo et al, 2004), in macrophages (Frankenberger et al, 2005) and in T cells (Barrat et al, 2002;Richards et al, 2000).…”

“…According to in-vitro studies GC can induce IL-10 in monocytes (Mozo et al, 2004) and macrophages (Frankenberger et al, 2005). Furthermore, GC treatment will induce IL-10 in T cells in therapeutic settings (Karagiannidis et al, 2004) and in in-vitro culture (Barrat et al, 2002;Richards et al, 2000).…”
