Glucose starvation increases V-ATPase assembly and activity in mammalian cells through AMP kinase and phosphatidylinositide 3-kinase/Akt signaling

Abstract: The vacuolar H-ATPase (V-ATPase) is an ATP-driven proton pump involved in many cellular processes. An important mechanism by which V-ATPase activity is controlled is the reversible assembly of its two domains, namely the peripheral V domain and the integral V domain. Although reversible assembly is conserved across all eukaryotic organisms, the signaling pathways controlling it have not been fully characterized. Here, we identify glucose starvation as a novel regulator of V-ATPase assembly in mammalian cells. … Show more

“…To understand how loss of WDR7, CCDC115, and TMEM199 resulted in expansion and over-acidification of the endolysosomal compartments, we extracted cytosolic and membranous proteins from the polyclonal KO cells and measured the relative abundance of the cytosolic V 1 A and transmembrane V 0 D domain subunits of the V-type ATPases via western blot 46 . We observed an enrichment of V 1 A subunit in the cytosolic fraction Fig.…”

“…Increased V‐ATPase assembly due to amino acid starvation may be due to increased intracellular levels of free amino acids as a result of increased protein degradation in the lysosome 20. PI3‐kinase‐dependent signaling appears to play a central role in inducing V‐ATPase assembly in response to increased glucose, but not amino‐acid deprivation 20, 21. V‐ATPase assembly is also promoted by interaction with aldolase or phosphofructokinase22, 23 and this may be important for AMPK activation 10…”

“…The response of V-ATPase to glucose depletion has been most thoroughly studied in yeast, and the mechanisms driving this response are still under investigation. In this issue, McGuire and Forgac (3) show that mammalian V-ATPase responds to glucose deprivation in a novel manner, highlighting fascinating species-specific differences in the ways that organisms respond to energy deficits.…”
