Glutaredoxin 2 promotes SP-1-dependent CSPG4 transcription and migration of wound healing NG2 glia and glioma cells: Enzymatic Taoism

Abstract: Redox regulation of specific cysteines via oxidoreductases of the thioredoxin family is increasingly being recognized as an important signaling pathway. Here, we demonstrate that the cytosolic isoform of the vertebrate-specific oxidoreductase Glutaredoxin 2 (Grx2c) regulates the redox state of the transcription factor SP-1 and thereby its binding affinity to both the promoter and an enhancer region of the CSPG4 gene encoding chondroitin sulfate proteoglycan nerve/glial antigen 2 (NG2). “…This activation also promotes the EMT process and the expression of VM related genes genes. Additionally, prior studies also report that CSPG4 may foster proliferation, migration, and invasion of tumor cells by regulating EMT-related pathways ( Winship et al, 2017 ; Wilms et al, 2022 ). To sum up, CAFs promote VM formation through multiple pathways including fibronectin and laminin as well as potentially key mechanisms such as HGF/MET and CSPG4 pathways.…”

“…Enzymatically active apo-Grx2 is required, for instance, for brain and vascular development (Bräutigam et al , 2011, 2013), and promotes cell migration of cancer cells (Gellert et al , 2020; Wilms et al , 2022). The same mode of cluster coordination was also demonstrated for the human monothiol Grxs 3 and 5 (Haunhorst et al , 2010; Johansson et al , 2011) that function in iron homeostasis and FeS cluster biosynthesis, as well as for various Grxs from other species, for overviews see for instance (Berndt & Lillig, 2017; Rouhier et al , 2010).…”

“…Moreover, the co-employment of irf8 −/− mutants lacking microglia can be useful in further investigating their the role in GBM progression and survival [140]. Zebrafish neural stem cells, astrocytes, and oligodendrocytes have also been defined by marking, respectively, the glial fibrillary acid protein (gfap) in Tg (gfap:GFP) [106] and oligodendrocyte transcription factor 2 (olig2) in Tg (olig2:GPF) [141] lines.…”
