Glycosphingolipid patterns in primary mouse kidney cultures

Abstract: Primary kidney cultures from C57BL/6J mice, 6 weeks of age or older, were produced using D-valine medium to select for epithelial cell growth. After allowing the cells to attach and proliferate for 1 week following plating, medium was changed once per week. Cells formed nearly confluent monolayers during the second week of culture. The cultured cells contained all of the glycosphingolipids seen in the adult kidney, analyzed by high performance liquid chromatography as their perbenzoyl derivatives. Glucosylcera… Show more

“…In contrast to rat tissues, mouse renal tissues contain appreciable concentrations of Gal(al-4)Gal,-containing glycolipids (7,328). The concentrationl of total glycolipids, and of each globoseries glycolipid, differs between different strains of mice and between male and female mice (7,328).…”

“…The concentrationl of total glycolipids, and of each globoseries glycolipid, differs between different strains of mice and between male and female mice (7,328). In female BALB/c mice, antiglobotriaose antibodies identify receptors for P fimbriae in the vagina, bladder, ureter, renal pelvis, collecting duct, and tubular cells, with no staining of the glomerulus or loop of Henle (391).…”

“…human, pig, rat, mouse, and hamster, Gal␣1-4Gal is normally found in the terminal and internal positions of glycolipids (1)(2)(3)(4)(5), as in P 1 (Gal␣1-4Gal␤1-4GlcNAc␤1-3Gal␤1-4Glc␤1-1Cer), P k (Gal␣1-4Gal␤1-4Glc␤1-1Cer) antigens, and disialosyl galactosyl globoside (NeuAc␣2-3Gal␤1-3(NeuAc␣2-3)GalNAc␤1-3Gal␣1-4Gal␤1-4Glc␤1-1Cer) (6 -8). The glycolipids containing Gal␣1-4Gal are known as targets on host cells for infections of some pathogenic microbes, e.g.…”

“…At the receptor level, the globoside isolated from the rat kidney contain a Gal  (1-3) Gal  linkage instead of the critical Gal (1-4) Gal  linkage present in humans [8]. In contrast, mouse renal tissue has been found to contain an appreciable concentration of Gal  (1-4) Gal  containing glycolipid which matches with that of humans [8]. Further, it has been recognized that Tamm-Horsfall glycoprotein (THP) present in mouse urine is physically, chemically and immunologically almost identical to THP found in human urine [9].…”

“…However, rat uroepithelial cells do not match human cells in the expression of receptors for P-fimbriae of Escherichia coli [7]. At the receptor level, the globoside isolated from the rat kidney contain a Gal  (1-3) Gal  linkage instead of the critical Gal (1-4) Gal  linkage present in humans [8].
