Glypican 4 regulates planar cell polarity of endoderm cells by controlling the localization of Cadherin 2

Abstract: Non-canonical Wnt/Planar Cell Polarity (Wnt/PCP) signaling has been implicated in endoderm morphogenesis. However, the underlying cellular and molecular mechanisms of this process are unclear. We found that during convergence and extension (C&E) in zebrafish, gut endodermal cells are polarized mediolaterally, with GFP-Vangl2 enriched at the anterior edges. Endoderm cell polarity is lost, and intercalation is impaired, in the absence of glypican 4 (gpc4), a heparan-sulfate proteoglycan that promotes Wnt/PCP… Show more

“…Previous studies performed on Zebrafish and Xenopus embryos have highlighted GPC4 function during ME convergence extension movements at gastrulation in which GPC4 controls polarity and directed migration of mesodermal cells 18,20,54,55 . Moreover, in Zebrafish embryos GPC4 promotes DE cell polarity by influencing localization of N-CAD on the cell surface through regulation of Rab5c-mediated endocytosis 56 . Future experimental settings will clarify whether mechanisms involving GPC4-mediated endocytosis of membrane proteins participating to cell-cell and cell-matrix contacts underlie the epithelial integrity defects found in GPC4sh hiPSCs.…”

“…Glypican-4 is a well-characterized cell surface heparan sulfate proteoglycan involved in the trafficking of morphogens, including Wnt and hedgehog, and plays a vital role in their development ( Song and Filmus, 2002 ). Glypican-4 has been shown to activate both the Wnt classical and PCP pathways ( Ohkawara et al, 2003 ; McGough et al, 2020 ; Balaraju et al, 2021 ). Based on our observations regarding actin cytoskeletal protein levels in Dex treated TM cells, it is plausible that GPC4 activates the Wnt/PCP pathway in response to Dex treatment.…”

“…Glypicans are heparan sulfate proteoglycans (HSPGs) attached to the extracellular surface of the plasma membrane through a glycosyl-phosphatidylinositol (GPI) anchor. [47][48][49] While there are six members of glypicans (GPC1-6), GPC3 is the most investigated one in the literature. 50 GPC3 plays a role in endocytosis and degradation of GPC3-Hedge-hog (Hh) Complex.…”
