Growth Hormone and Prolactin Secretion after Metoclopramide Administration (DA2 Receptor Blockade) in Fertile Women*

Abstract: In this report, we will describe the results of a cross-sectional study to assess PRL and GH secretion during the early follicular phase in 22 fertile patients after metoclopramide administration in order to achieve a dopaminergic DA2 receptor blockade. Blood samples were collected at - 15, 0, 15, 30, 45 and 60 minutes. PRL, GH, estradiol, IGF-I, TSH, glucose, and insulin were measured in the samples taken at - 15 and 0 minutes. The existence of a correlation between GH and PRL secretion was investigated. All … Show more

“…Endometriosis is a heterogeneous disease associated with infertility and one of the main causes of IVF. Furthermore, several mechanisms involving granulosa cells were described for this group of patients, involving prolactin secretion, decreased anti-Mullerian hormone and dysfunction in BMP-6 and SMAD4 in cumulus cells linked to peritoneal endometriosis ( Cunha-Filho et al. , 2001 ; 2003 ; De Conto et al, 2017 ; 2020).…”

Furthermore, several mechanisms involving granulosa cells were described for this group of patients, involving prolactin secretion, decreased anti-Mullerian hormone and dysfunction in BMP-6 and SMAD4 in cumulus cells linked to peritoneal endometriosis (Cunha-Filho et al, 2001;2003;De Conto et al, 2017;. Nowadays the effect of endometriosis during an IVF cycle is disputed and has recently been questioned by several authors, but it is essential to understand the role of this disease during the IVF process to enhance pregnancy rates (Opøien et al, 2012;Barbosa et al, 2014;González-Comadran et al, 2017;Caran et al, 2021).…”

“…Data on the effect of metoclopramide on GH secretion are mixed. Two previous studies have shown no impact on GH levels post‐metoclopramide, 22,23 whilst two other studies report metoclopramide may stimulate GH secretion including one study of adolescent males of short stature 24 and another in patients with diabetes mellitus on insulin therapy 25 . However, the exact mechanism for this proposed stimulation in both studies was unclear.…”

“…It will be interesting to study whether other hormones that have been associated with SLE activity and whose secretion is also induced by metoclopramide, such as growth hormone ( 36 , 37 ), and estrogens ( 38 , 39 ) could also influence the formation of GC.…”
